Specimen Requirements


Patient Preparation
  • Cleanse the area by flushing with sterile physiological saline.
Specimen Collection
  • Clinical Specimens

-Approved specimen source: Ulcer material from margins of genital lesions, lymph node aspirates, pus, aspirates from buboes.

-Material should be collected from the base of the ulcer with a swab.

-Place swab in transport such as Amies or Amies with charcoal.

  • Isolates

-Must be grown on Chocolate agar or Blood Agar Plates for Haemophilus ducreyi (BAHD)

Specimen Labeling
  • Specimen must be labeled with at least two unique patient identifiers and match accompanying PH-4182 Clinical Submission Requisition.
Specimen Processing
  • None
Specimen Storage and Preservation
  • Place in transport such as Amies or Amies with charcoal. Refrigerate at 2-8°C and transport specimen within 4 hours of collection.
  • Isolates should be stored at ambient temperature.
Specimen Transport
  • Ship clinical specimens at 2-8°C on ice packs overnight within 4 hours of collection.
  • Ship isolates at ambient temperature.
  • All infectious substance shipments must conform to U.S. Department of Transportation (DOT) Hazardous Materials Regulations (HMR 49 C.F.R. Parts 171-180). 
Specimen Acceptability and Rejection
  • Specimens that do not meet all specimen requirements will be rejected.
